I once had a tire blow out 12 hours away from home. I'm pretty picky on
tires and didn't have much choice on where to get a replacement. They were
trying to sell me on getting 4 new tires so I didn't break the AWD, but I
said I was pretty sure I was ok because the wheels are almost always
turning slightly different speeds because wear and not going exactly
straight, etc...

But, I called Audi Of America's hotline, and they confirmed it:

As long as you are talking the same size tire, differences in wear will
not damage the Quattro system. So, if you have 3x 220 40 R 18 tires on
it and you put a new 220 40 R 18 tire on it, you're fine. DO NOT put a
   220 35 R 18 on it, for example.

That's what Audi told me. The exact car in question was a 1997 A4 2.8Q
(which didn't have the above tire sizes on it, I don't remember what they
were, the above are closer to what my S4 wears, I just replaced two of
them).
