5000TQ trany noise under lock

Last week i tried the lock position, just to see if everything is ok. Was ok.

Yesturday i had a front flat tire. I was pulling an heavy trailer (wood filled).

I was on the side of the road when i lock the center lock to get better grip, cause the road there was no very good...

When i began to run the car i heard loud vibration come out the tranny and its like i run with brake apply !!!!! at 50km/h the vibration is for sure not normal...

Why last week everything was fine and now after a flat front tire the thing got bad ?  i mesured the spare tire , its 2feet high from the ground, same as the 3 other wheels with 16 inchs tires. I noticed the vibration was much more loud with another smaller spare tire,,, i understand the wheels dont turn at the same speed cause the spare is to much diff from the others... but i dont went i use the right spare.... 

i ll replace the bad tire and see what ll happens.

Any good advice ? tought ?
