Yes, it will work.  You will also pressurize the crankcase... so a good seal of your oil cap is needed, and you need to make sure you don't blow out your dipstick... if you want to hear potentially unwanted leaks.

I am going to pressure test my charge piping and wondered if its possible to 
pressure test the intake manifold at the same time. A friend with a 91 200 says 
he just hooks his tester on the MAF hose and pumps it up to 15 psi then unscrews 
the oil cap to relieve pressure. Will this really work?
