That would be a "no-no."  Clamping the hose adds wear and can readily brake
down the hosing reinforcement.  If you have that much dirty brake fluid in
the hose, then you've got a systemic problem.  That problem is generally
"water logged" fluid (brake fluid is hygroscopic) due to not enough brake
fluid changing.

If you think the car hasn't had regular MAINTENANCE, than a better approach
is to open the bleeder valve before you retract the piston.  The dirty fluid
will come out there rather than go back upstream (path of least
hydraulic resistance in this case).  Never clamp a brake line.
