The glovebox switch only works with the ignition on (so some lowlife can't break into your car and gain trunk access).
Give this a try, if you haven't yet.

Once the trunk is open, remove the screws holding the inside trim panel and, most likely, the actuator arm has become
disconnected from the lock. Pop 'er back in and you're good to go. BTW, my 2004 S4 has a glow-in-the-dark trunk release
handle on the inside of the trunk. Think it's there for the yet-to-be-announced Trunk Monkey option.
